Comparison of results by four different procedures for determination of 17-hydroxycorticosteroids.

چکیده
A Porter and Silber procedure, a fluorometric procedure, a double-isotope derivative procedure, and a competitive protein-binding assay procedure were used to determine 17-hydroxycorticosteroids in plasma from normal individuals and from patients with endocrinopathies. Results from each of these procedures were intercompared. Results of the fluorometric procedure and the competitive proteinbinding assay compared favorably with results obtained by the more elaborate and difficult doubleisotope derivative method. The Porter and Silber method is less specific. This study should be a useful aid in comparing values with those of other laboratories and in selecting the most advantageous method for use by a particular laboratory.

Cefoxitin interferes with the "Clini-Skreen" column method for urinary 17-hydroxycorticosteroids.
Cefoxitin interferes with determination of urinary 17-hydroxycorticosteroids. The apparent concentration of hormone is increased from three- to 10-fold in samples from patients receiving cefoxitin when the Amberlite XAD-2 "Clini-Skreen" column is used. To determine the mechanism of interference, we reacted aqueous solutions of cefoxitin, cortisol, cortisone, and 11-deoxycortisol with phenylhydr...
متن کاملThe use of tetramethylammonium hydroxide in the Zimmermann reaction.
The use of tetramethylammonium hydroxide (T.M.A.H.) in place of potassium hydroxide in the Zimmermann reaction has been investigated. Although various pure steroids have different colour equivalents, a comparison of the results of 17-ketosteroid and 17-hydroxycorticosteroid estimations on a series of urines showed that the differences for the two reagents were only small and may be ignored. Tet...
متن کاملA Modified Procedure for the Estimation of Total 17-hydroxycorticosteroids (porter-silber Chromogens) in Urine.
A modified procedure for the determination of urinary 17-hydroxycorticosteroids(dihydroxy-acetonyl-corticoids) is described. The use of ethyl ether in a single extraction step and the stabilization of the phenylhydrazine-sulfuric acid reagent with sodium chloride are the unique features of the method. Studies of chromogenicities of steroids, recovery, pH effect on extraction, and salt effect on...

hydrochloride (10 mg three times a day for as long as 20 days) did not affect values for 17-hydroxycorticosteroids. In vitro data do show that there is a small increase in the apparent value for urinary 17-hydroxycorticosteroids when chlordiazepoxide or its lactam or N-despiethyl metabolite is added to urine. However, this increase does not appear to be large enough to be of significance. The f...
